Expedition onboard the R/V “Papanin” on August 11-13, 2024

On August 11-13, 2024, within the RSF project No. 23-14-00028 “Microeukaryote communities in the Angara Cascade reservoirs” (led by Ye.V. Likhoshway), a water sampling expedition was carried out onboard the R/V ‘Papanin’ to determine the species diversity and structure of microeukaryotes in the photic layer of the southern basin of Lake Baikal (SB: 3 km from Listvyanka) and Irkutsk reservoir (IR: 8 stations, including Kurminsky, Elovy, and Ershovsky bays). Water transparency was determined with a Secchi disc. The pH, temperature and EC were measured in the water. Water samples were collected on filters for examination by scanning electron microscopy, quantification of phytoplankton composition by light microscopy, DNA extraction for sequencing, and determination of microeukaryote community composition. Phytoplankton samples were collected with a Juday net to isolate and culture microalgae. As a result of the expedition, the temperature of the upper water layer (0.5 m) was found to be up to 19°C in SB and up to 21°C in IR. The pH of the upper water layer was 8.34 in SB and 8.36-8.66 in IR. Preliminary analysis of selected samples by light microscopy revealed that the phytoplankton of SB and IR is represented by diatoms, chrysophytes, and green microalgae. Phytoplankton in the bays and the central part of the reservoir were similar. The species composition of phytoplankton was represented by diatoms Asterionella formosa Hassall, Aulacoseira granulata (Ehrenberg) Simonsen, Nitzschia graciliformis Lange-Bertalot & Simonsen, green microalgae Monoraphidium griffithii (Berkeley) Komárková-Legnerov, dinophyte Ceratium hirundinella (O.F.Müller) Dujardin and chrysophytes Dinobryon cylindricum O.E. Imhof., D. divergens O.E. Imhof, D. sociale (Ehrenberg) Ehrenberg.
